The chromosomes of sperm come from the male's own genes and are divided into X and Y. We use a microscope to examine the sperm chromosomes and then analyze the sperm chromosomes to help us prevent the occurrence of Down syndrome. The sperm test items are to check the volume of semen and the number of sperm. It is also necessary to check the survival rate and pH of the sperm, mainly to allow the sperm to enter the woman's fallopian tube and combine with the egg.

Sperm chromosome examination is a diagnostic method that refers to the direct analysis of sperm chromosomes. Including sperm capacitation treatment, golden hamster superovulation, in vitro fertilization, sperm chromosome preparation and chromosome analysis and other processes. First, golden hamster eggs obtained from superovulation are treated with hyaluronidase to digest the granulosa cells on the periphery of the eggs, and the zona pellucida is digested with trypsin. They are then incubated with pre-capacitated human sperm for 3 hours, with a sperm concentration of 1 to 2 × 107/ml.

After the sperm and eggs are combined, the eggs are washed several times with BWW culture medium to remove the surface-adhered sperm that have not entered the eggs. The eggs are continued to incubate for 12 hours. Finally, the eggs are treated with 1% sodium citrate solution for 10 to 15 minutes. After a stereomicroscope shows that the eggs have fully expanded, the eggs are sucked out, placed on a clean glass slide, fixed with 3:1 methanol glacial acetic acid solution, dried at room temperature, stained with Giamsa solution, or subjected to various banding treatments.

    Routine inspection items

    1. Semen volume

It is necessary to check the total amount of semen discharged by male friends in one ejaculation. Under normal circumstances, normal men can discharge 1.5 ml to 6.0 ml.

    2. Sperm number (concentration or density)

It is necessary to check the number of sperm per milliliter in the male friend's semen. Under normal circumstances, the number of sperm in male semen is 20 million/ml.

    3. Sperm survival rate

It is necessary to check the motile sperm in the male friend's sperm. In the normal male friend's semen, there should be at least 60% viable sperm per 100 sperm.

    4. pH value (PH value)

It is also necessary to check the pH of sperm. Normally, the pH of male sperm should be maintained between 7.0 and 7.8. If the pH of sperm is 7 or 8, it means that the motility of male sperm has dropped significantly.

    5. Color

Normal male semen is milky white or off-white. If the semen is yellow-green, it means that the male has an inflammatory infection of the reproductive tract.
